Until recently the transporters responsible remained obscure, but a breakthrough occurred with the discovery of the SLC26 transporters family. It is now clear that the SLC26 transporters have broad physiological functions since mutations in several members are linked to a variety of diseases. This book describes the properties of this family in detail, with contributions from the leading global researchers in the field. Complementary views from experts on other ion channels are offered in the discussions, which make fascinating reading. This family consists of at least 10 genes, each of which has several splice variants. Most members of the family are expressed in the luminal membrane of epithelial cells. Characterization of anion transport by three members has revealed that all function as Cl-/HCO3- exchangers, suggesting that SLC26 transporters are responsible for the luminal Cl-/HCO3- exchange activity. The SLC26 transporters are activated by the CF transmembrane conductance regulator and activate it in turn, leading to a model in which these molecules act together to mediate epithelial Cl- absorption and HCO3- secretion. The book includes chapters on the transport of other molecules by the SLC26 family, including oxalate in the kidney and sugars in cochlear hair cells amongst others. It also describes recent discoveries that most SLC26 transporters bind to scaffold proteins and that they all contain a conserved domain predicted to participate in protein-protein interactions. These suggest the SLC26 transporters exist in complexes with other Cl- and HCO3- transporters, and possibly other regulatory proteins. This book explores the functional role of these interactions, leading to better understanding of transepithelial fluid and electrolyte secretion and the diseases associated with it.

Under this Act, any scientific procedure carried out on any living vertebrate animal, and one species of octopus, which is likely to cause pain, suffering, distress or lasting harm is a regulated procedure requiring licence authority. The data is structured to comply with EU requirements, although it is far more extensive than required by Europe. In 2003, there were 2.79 million scientific procedures conducted, an increase of 2.2 per cent on the previous year. Most experiments and tests were conducted on mice, rats and other rodents (85 per cent), with the remainder on birds (four per cent) and fish (six per cent). Dogs, cats, horses and non-human primates (usually macaques and marmosets) combined amounted to less than one per cent of procedures. Genetically modified animals were used in 27 per cent of cases, an increase of one per cent on 2002 and 19 per cent on 1995). Non-toxicological procedures accounted for 84 per cent of procedures, with the main areas of use being for immunological studies, pharmaceutical research and development, and cancer research.

My interest always vanished when I was confronted with complex data pointing to func tional redundancy of cytokines in cell culture systems. When gene targeting in the mouse germline became possible, this frustration came to an end. We and others immediately embarked on analyzing the in vivo function of cytokines and the problem of functional redundancy with this powerful new approach. The early cytokine gene knockouts performed by colleagues in Wiirzburg (IL-2) and by ourselves (IL-4 and IL-l 0) seemed to give clear answers and at the same time led to surprises: Each of these cytokines apparently had its own special and irreplaceable function, and this function could be quite distinct from what had been anticipated from functional experiments in vitro. Al though the latter finding is of course a wonderful incentive for fur ther research, the former is pleasing in a general sense since it highlights the value of each of those one hundred thousand genes or so in our genome, cherished by evolution to become respectable mem bers of the community. Even in the present era of "genomics" there will be no way around the careful functional analysis of each gene by itself.
